You can if you want, but there's really no need.
The old reason to keep backups of the original coding file was to have a copy that you could revert back to factory settings in case of problems.
That's not necessary any more as there is a function in E-Sys to reset an ECU (or all ECU's) to factory settings without any saved files. It's called VO coding and is done when adding/removing option codes from the car.
